【Verification principle】
Pathogenic Staphylococcus can produce plasma coagulase, which turns fibrinogen in plasma into insoluble fibrin and attaches to the surface of bacteria, forming clots.
【Composition】
Rabbit plasma, citrate.
【Usage method】
Add 0.5ml of matching solvent to each penicillin bottle, gently shake until completely dissolved, then add 0.3ml of fresh broth culture, gently rotate the bottle until fully mixed, and place it at 36 ± 1 ℃ for cultivation. Observe the results every half hour until the end of 6 hours.
【Inspection results】
When tilted or inverted, if a solidified block appears, or if the volume of the solidified block is greater than half of the original volume, it is considered positive. Staphylococcus aureus is positive and plasma clots; Staphylococcus epidermidis is negative and the plasma does not coagulate.
【Note】
When observing the results, it is important to avoid forceful vibration of the penicillin bottle to prevent the clot from shattering.
