【Verification principle】
Adsorbing a certain amount of nutrients such as lactose, bromocresol violet, and TTC onto paper, the coliform bacteria can ferment lactose in the paper, produce acid, and lower the pH value. The bromocresol violet indicator changes from purple to yellow. At the same time, TTC is reduced by the Escherichia coli group to form easily recognizable red colonies, which can display red spots (or halos) on the yellow background after acid production.
【Composition】
Made from lactose bile salt culture medium, TTC and other raw materials adsorbed on paper.【Usage method】
After pre-treatment according to industry regulations, the sample to be tested is coated on paper, placed in a sterile plastic bag, and incubated at 35 ℃ to 37 ℃ for 18-24 hours to observe the color changes of the paper.
【Inspection results】
If the paper turns yellow and shows red spots or patches of red halo on the yellowed background, it is considered positive for coliform bacteria.
If the paper remains purple or shows red spots on a purple background, but there is no yellow around it, it is negative for coliform bacteria.
【Note】
When picking up paper pieces, sterile tweezers should be used to slowly remove them to avoid damage.
